Job description

The Position

The ADME Chapter generates data from in vitro and in vivo studies on target compounds to support translation from early research to development across all therapeutic modalities in the Roche pRED portfolio. 

We use a variety of state-of-the-art validated cellular/subcellular models, bioanalytical techniques and automation to characterize compound drug metabolism and pharmacokinetic properties in compliance with global quality and regulatory requirements. The data we generate is crucial to the timely delivery of safe, effective and high-quality therapies to patients.

Our team is located in Basel and currently comprises 8 full-time employees and 1 PhD student. Our focus area is in enzyme-mediated drug-drug (DDI) interaction risk assessment through determination of metabolic enzyme inhibition, induction as well as enzyme phenotyping. 

Your tasks and responsibilities:

  • You will perform in vitro assays to characterize metabolism of probe CYP450 substrates using hepatic microsomes and individual recombinant enzymes

  • You will generate in vitro data to correlate metabolic activity of selected recombinant and hepatic CYP450 enzymes using LC-MS and/or radio-HPLC

  • You will verify a variety of CYP450 chemical inhibitors and probe substrates against recombinant enzyme data

  • You will help to validate an automated platform for in vitro enzyme phenotyping assessment

  • You will assist in gathering data for improved PBPK modeling prediction 

Your profile:

  • You are currently a MSc/PhD student in chemistry, biochemistry, pharmacokinetics or equivalent discipline with an interest in drug metabolism, pharmacokinetics and pharmacodynamics

  • You have a strong desire to expand your knowledge of metabolic enzyme biochemistry, kinetics assessment and the respective experimental designs with the help of automation.

  • You are interested or have some experience in bioanalytical method development/validation using radio-HPLC or LC-MS/MS

  • ​You wish to explore translational approaches leveraging in vitro data and clinical data using physiologically-based pharmacokinetic (PBPK) modeling and understanding the importance of ‘victim’ DDI risk assessment in drug development.

  • You have strong communication skills and the ability to work in a fast-paced and quickly changing environment in close collaboration with partner functions. Proficiency in English is required.

The internship will start as soon as possible.
