BioAnalytical Sciences (BAS) develops, implements and provides strategic direction for state of the art bioanalytical methods and automation systems. Our data and expertise enable the characterization of the pharmacokinetic, immunogenic and in vitro biological activities of Genentech biotherapeutics during drug development and throughout the product life cycle.

The experimental work involves continuing the development of an imaging method to detect ADCC in real time. Target cells will be opsonized with a panel of test antibodies (both positive and negative controls) and plated. NK92 cells will serve as the effector cells. As the NK92 cells target and lyse the target cells, the imaging system will capture the cell lysis. The timing of the target cell lysis will be calculated over time and converted into a kinetic curve. Cell lysis will be analyzed using multiple readouts. One approach is to label the target cells with a cell permeant dye. As the target cell is lysed by the NK cells, the dye is released and the loss of fluorescence in the target cell can be measured on the cell imaging system. A second approach is to utilize DAPI incorporation. As the target cell is targeted for lysis by the NK cell, it starts to undergo apoptosis. DAPI dye can then integrate into the nucleus, marking the target cell as undergoing lysis. By measuring the increasing number of DAPI positive target cells, it will be possible to measure the number of cells undergoing lysis.
