Description de l'offre

The group of "Chemistry for Recognition and Study of Biological Assemblies" (CREAB) at SyMMES (UMR5819, CEA Grenoble, France) researches candidates for a 6-month training period beginning early 2019. The trainee will work on the PLANARPORE project, financed by the ANR and putting together three laboratories: SyMMES/CREAB, LAAS (Toulouse, France) and IAB (Grenoble, France).

The health of an individual depends on the progress of her/his immune response which organizes the defenses of the body against infections, inflammatory and autoimmune diseases, elimination of cancer cells, graft rejections. The PLANARPORE project aims to design a chip analysis platform for the precise definition of the functions of immune cells (lymphocytes) including their secretions. In this project, we propose to use a two-dimensional (2D) pore technology in order to trap cells to analyze. These 2D pores are based on the restriction of a microfluidic channel. Their functionalization with biomolecules will be highly localized and will be obtained by conventional techniques such as spotting or by a more innovative method inspired from electrochemistry (CLEF technique developed in 2009). The detection technique for the different biological objects (electrical or fluorescence) will also be examined. The design of multi-scale biosensors (combined detection of cells and their secretions) will then be achieved on the basis of the 2D technology through the manufacturing of chips displaying a succession of pores of different sizes.

The trainee who will join the SyMMES/CREAB will, at first, be in charge of the biofunctionalization of chips comprising a single pore. She/He will then study the handling of cells in flow on these chips and their detection in fluorescence or electrically. Her/His work will be highly multidisciplinary at the interfaces between chemistry, electrochemistry, microfluidics and biology. The applicant shall have a motivation for these four scientific fields.
